Engineering a minimalist protein scaffold for macromolecular recognition

Abstract:

Although monoclonal antibodies are incredibly useful molecules for binding targets in disease or for analytical biochemistry, there are some applications for which a smaller and/or more robust and portable modular protein domain are advantageous. Dr. Wittrup will discuss his team's efforts to describe a hyperthermostable protein of about 60 amino acids (1/20th the size of an antibody) for antibody-like binding properties. He’ll also present data on application of such small, stable binders to highly multispecific anti-receptor antibodies and cytoplasmic drug targeting.
